The role of xylulose 5-phosphate in xylose metabolism of Lactobacillus pentosus.

Lampen (1) observed that cell-free extracts of LactobaciZZus pentosus grown on D-xylose catalyzed the disappearance of n-xylose in the presence of ATP.’ The phosphate esters which accumulated were identified as a mixture of D-ribose 5-phosphate and D-ribulose 5-phosphate. Neither n-xylose 5-phosphate nor D-xylulose 5-phosphate was found in the reaction mixture. Mitsuhashi and Lampen (2) also demonstrated the presence of xylose isomerase in these extracts and, on the basis of the fact that D-xylulose was esterified more rapidly than D-xylose, it was postulated that the following reaction sequence takes place.

The inability of two Lactobacillus strains to ferment D-xylose was complemented by the introduction of Lactobacillus pentosus genes encoding D-xylose isomerase, D-xylulose kinase, and a D-xylose catabolism regulatory protein. This result opens the possibility of using D-xylose fermentation as a food-grade selection marker for Lactobacillus spp.
